I'm extracting protein from raw mammalian spleen and would like to use protease inhibitors. I have six of them, but don't know which one to use. Any thoughts?
EDTA, PMSF, Benzomidine HCl, N-ethylmalamide, pepstatin A, and aproteinin.
Answer:
first, you need to know what kind of protein you're trying to purify - whether it contains metal ions, if it's a serine protease etc. then you decide which protein inhibitors to use - e.g. EDTA will complex metal ions so you can't use it to purify metalloproteins, PMSF will react with active site serine, etc. You can also use protease inhibitor tablets or cocktails that are specifically designed for a specific type of cells, check with Sigma or Roche for the content
